ABSTRACT: Cling wrap has properties similar to Tegaderm™, an expensive band-aid that
has been used as a transport membrane of keratinocytes. PRP gel and fibrin glues are
proved to be useful as the cellular adhesive on cellular transplantation. We
hypothesized that either PRP gel or fibrin glues can enhance plastic-keratinocytes
adhesion and can be used as a replacement material for Tegaderm™.
In order to have an evidence of our hypothesis, an invitro study was designed
to determine passage 2 human keratinocytes adhesion on cling wrap and compared to
Tegaderm™. Forty micro liters of 1 x 105 keratinocytes/mL were dropped onto cling
wraps and Tegaderm™ either coated by fibrin glues or PRP gel. Measurement of
keratinocytes adhesion was using the MTT-assay, performed after 72 hours
incubation, and read by 570 nm of spectrometer. Improving of research validity was
performed by eight times repeated procedures. The means of optical density of blue
formazan among various groups were analyzed using the one-way Anova and
Kruskal Wallis test with post hoc analysis by Mann-Whitney test.
The results showed adhesion of keratinocytes on uncoated cling wrap better
than uncoated Tegaderm™ (P<0,05). Fibrin glue coating on the cling wrap
significantly (P<0.05) enhanced the adhesion of keratinocytes. This adhesion is equal
to fibrin glue coated Tegaderm ™. This fact is not found among PRP gel coated cling
wrap. Conclusion, cling wrap can be used as keratinocyte transport membrane in
cellular grafting
INTISARI: Plastik cling wrap bersifat mirip Tegaderm™, plester pembalut luka yang biasa
digunakan sebagai wahana pembawa keratinosit, meskipun dipandang mahal. Gel
PRP dan lem fibrin merupakan bahan adhesif yang biasa digunakan dalam
transplantasi selular. Penambahan lem fibrin dan gel PRP pada plastik cling wrap
diharapkan dapat mengganti fungsi Tegaderm™.
Untuk membuktikan dugaan diatas, dirancang penelitian invitro dengan
menilai adhesi 40 μL suspensi keratinosit passage 2 berdensitas 1 x 105 sel/mL yang
diteteskan pada plastik cling wrap masing-masing berlapis lem fibrin dan gel PRP,
dan membandingkannya dengan Tegaderm™. Untuk meningkatkan validitas
penelitian, pengulangan dilakukan sebanyak delapan kali. Pengukuran adhesi
keratinosit dilakukan setelah inkubasi selama 72 jam, menggunakan MTT-assay dan
dibaca dengan spektroskopi 570 nm. Rerata densitas optis formasan biru dianalisa
dengan uji one-way Anova dan uji Kruskal Wallis dengan analisis post hoc uji Mann-
Whitney. Hasil penelitian menunjukkan adhesi keratinosit pada plastik cling wrap tanpa
lem lebih baik dibandingkan Tegaderm™ tanpa lem (P<0,05). Pelapisan lem fibrin
pada plastik cling wrap meningkatkan adhesi keratinosit secara bermakna (P<0,05),
setara dengan adhesi keratinosit pada Tegaderm™ berlem fibrin. Hal serupa tidak
terjadi pada pelapisan plastik cling wrap dengan gel PRP. Kesimpulan, plastik cling
wrap dapat digunakan sebagai wahana pembawa keratinosit dalam grafting selular.
